Agarose

Agarose is a polysaccharide formed by the union of β-D-galactopyranose and 3,6-anhydro-α-L-galactopyranose linked by 1-4 bonds. This basic repeating unit, called agarobiose, forms long chains. This structure contains charged groups, such as sulfate and pyruvate, and uncharged groups, all of which are responsible for many of the properties of agarose. These properties can be adjusted to meet specific needs through the careful selection of raw materials.

Some important properties of agarose are:

  • Gel strength: Force necessary to break a gel, expressed in g/cm2 .
  • Gelling temperature: Temperature at which an agarose solution forms a rigid gel.
  • Melt temperature: Temperature at which the gel becomes a solution.
  • EEO (electroendosmosis): Measure of the number of charged groups present in the agarose.

D3 agarose has a very high molecular weight, much higher than other agarose, a characteristic that results from extremely high gel strength. The gel structure and exclusion limit are similar to D1 agarose.

D5

D500

Linear polymer with a very high molecular weight, giving gel structures unlike those of traditional agaroses. This characteristic, added to a very low sulphate content, produces a strong intercatenary interaction, which generates a gel with a very high gel resistance and a high exclusion limit.

Biomax is an ideal agarose for the rapid routine separation of DNA and RNA fragments, as well as for PCR products, plasmid preparation and for detection, cloning and transfer techniques.

Low melting point (LM) agaroses are obtained by organic synthesis. The main properties of these agaroses are their low melting and gelling temperatures compared to standard agaroses. The low melting temperature allows the recovery of intact and biologically active nucleic acids, not damaged by the temperature below the denaturation temperature. The low gelling temperature ensures that the agarose will be in a liquid state in a temperature range in which in-gel manipulations can be performed without prior DNA extraction.

Characteristics:

  • Less gel strength than standard agaroses. Gels can nevertheless be easily handled.
  • Lower gelling/melting temperatures than standard agarose.
  • Greater clarity (gel transparency) than gels prepared with standard agarose. 
  • Increased screening capacity.

Nugel 3:1 Agarose

Suitable for separation of fragments 100 bp-1 kb. Form strong gels and easy to handle. Recommended for PCR products separation. Suitable for blotting of medium size fragments, 10-1000 bp.

These agaroses have been chemically modified to obtain a structure that improves the separation of small DNA fragments and the resolution between fragments of very similar size.

Characteristics:

  • High resolution capacity, in some cases close to that obtained with polyacrylamide gels.

  • Very clear and transparent gels, even at high concentrations.

  • Low viscosity solutions at high concentrations, allowing easy gel preparation at high concentrations (>4%) which allows the excellent separation of smaller fragment sizes.
